You can also go with Easy-Nightmare 1st and Infinity+Calamity on 2nd. It's recommended to go with Calamity first time and Infinity+Easy/Normal/Hard for second walkthrough if you go for trophies. It is designed for NG+, so there is no way to turn it on for your first walkthrough. It's an additional mode which adds 60 levels to all Greeds in game.

You can start with it and lower it later if it's too much, but you won't be able to turn Calamity on back after that.

Calamity is the hardest difficulty option. If you go for the trophies you need to finish it on Calamity once and once with Infinity mode. Even if you know them first two-three bosses will be the most difficult as you're still learning the game. If you're not familiar with Ys games battle system will be weird for you.
